Alterations in degree centrality and functional connectivity in tension-type headache: a resting-state fMRI study.

Previous studies have provided evidence of structural and functional changes in the brains of patients with tension-type headache (TTH). However, investigations of functional connectivity alterations in TTH have been inconclusive. The present study aimed to investigate abnormal intrinsic functional connectivity patterns in patients with TTH through the voxel-wise degree centrality (DC) method as well as functional connectivity (FC) analysis. A total of 33 patients with TTH and 30 healthy controls (HCs) underwent resting-state functional magnetic resonance imaging (rs-fMRI) scanning and were enrolled in the final study. The voxel-wise DC method was performed to quantify abnormalities in the local functional connectivity hubs. Nodes with abnormal DC were used as seeds for further FC analysis to evaluate alterations in functional connectivity patterns. In addition, correlational analyses were performed between abnormal DC and FC values and clinical features. Compared with HCs, patients with TTH had higher DC values in the left middle temporal gyrus (MTG.L) and lower DC values in the left anterior cingulate and paracingulate gyri (ACG.L) (GRF, voxel-wise p < 0.05, cluster-wise p < 0.05, two-tailed). Seed-based FC analyses revealed that patients with TTH showed greater connections between ACG.L and the right cerebellum lobule IX (CR-IX.R), and smaller connections between ACG.L and ACG.L. The MTG.L showed increased FC with the ACG.L, and decreased FC with the right caudate nucleus (CAU.R) and left precuneus (PCUN.L) (GRF, voxel-wise p < 0.05, cluster-wise p < 0.05, two-tailed). Additionally, the DC value of the MTG.L was negatively correlated with the DASS-depression score (p = 0.046, r=-0.350). This preliminary study provides important insights into the pathophysiological mechanisms of TTH.

Altered voxel-wise degree centrality associated with patients with comitant exotropia.

Objective of the study is to investigate the altered intrinsic functional hubs in patients with comitant exotropia (CE) using the voxel-wise degree centrality (DC) analysis method. A total of 28 CE patients and 28 healthy controls (HCs) similarly matched in sex, age, and education level were recruited in this study. All subjects underwent a resting-state functional MRI scan, the voxel-wise DC method was applied to evaluate brain network hubs alterations in CE patients. Then, the DC maps between two groups were chosen to be classification features to distinguish patients with CE from HCs based on the support vector machine (SVM) model. The algorithm performance was evaluated by a permutation test. Compared with HCs, CE patients exhibited significant enhanced DC value in the left cerebelum 8 and the right cerebelum 3; and remarkably decreased DC value in the right precentral gyrus, right anterior cingulated, and paracingulate gyri (two-tailed, voxel level: P < 0.01; GRF correction, cluster level: P < 0.05). However, no relationship was found between the observed average DC of the different brain regions and the clinical features ( P > 0.05). In addition, the SVM model showed an accuracy of 83.93% to clarify CE patients from HCs using the DC maps as a classification feature. CE patients displayed altered functional network hubs in multiple brain areas associated with cognition and motor control, and the DC variability could classify patients from HCs with high accuracy. These findings may assist to understand the neuropathological mechanism for the disease.

Resting-state functional magnetic resonance study of primary open-angle glaucoma based on voxelwise brain network degree centrality

ObjectiveTo investigate alterations in the functional brain networks of patients with primary open-angle glaucoma (POAG) by using the resting-state functional magnetic resonance imaging (fMRI) voxelwise degree centrality (DC) method. Materials and methodsThirteen patients with POAG and thirteen healthy subjects were recruited for this study, and each participant underwent a rs-fMRI scan. The voxelwise DC method was used to assess the features of spontaneous brain activity. The differences in the mean DC across brain regions between the POAG group and the healthy control group were analyzed, and the correlations between the DC values of altered brain regions and various clinical ophthalmic parameters were analyzed in the POAG group. ResultsCompared with healthy controls, patients with POAG exhibited significantly decreased DC values of the left superior frontal gyrus and the left postcentral gyrus as well as significantly increased DC values of the left superior occipital gyrus. In POAG patients, the DC value of the left superior occipital gyrus was significantly positively correlated with age (r = 0.571, P = 0.042) and negatively correlated with the intraocular pressure of the right eye (r=-0.625, P = 0.022). The DC value of the left superior frontal gyrus was significantly positively correlated with the right eye average cup-to-disc ratio (r = 0.683, P = 0.010), vertical cup-to-disc ratio (r = 0.779, P = 0.002), and pattern standard deviation (r = 0.567, P = 0.043). ConclusionThe results showed that altered DC values in three brain regions may reflect the underlying pathological mechanisms of POAG. Decreased DC values of the left superior occipital gyrus could be useful imaging markers for determining the extent of brain damage in POAG patients compared to healthy subjects.

Altered brain network centrality in patients with trigeminal neuralgia: a resting-state fMRI study.

Neuroimaging studies revealed that trigeminal neuralgia was related to alternations in brain anatomical function and regional function. However, the functional characteristics of network organization in the whole brain is unknown. The aim of the present study was to analyze potential functional network brain-activity changes and their relationships with clinical features in patients with trigeminal neuralgia via the voxel-wise degree centrality method. This study involved a total of 28 trigeminal neuralgia patients (12 men, 16 women) and 28 healthy controls matched in sex, age, and education. Spontaneous brain activity was evaluated by degree centrality. Correlation analysis was used to examine the correlations between behavioral performance and average degree centrality values in several brain regions. Compared with healthy controls, trigeminal neuralgia patients had significantly higher degree centrality values in the right lingual gyrus, right postcentral gyrus, left paracentral lobule, and bilateral inferior cerebellum. Receiver operative characteristic curve analysis of each brain region confirmed excellent accuracy of the areas under the curve. There was a positive correlation between the mean degree centrality value of the right postcentral gyrus and VAS score (r = 0.885, P < 0.001). Trigeminal neuralgia causes abnormal brain network activity in multiple brain regions, which may be related to underlying disease mechanisms.

Abnormal resting-state functional network centrality in patients with high myopia: evidence from a voxel-wise degree centrality analysis.

To investigate the functional networks underlying the brain-activity changes of patients with high myopia using the voxel-wise degree centrality (DC) method. In total, 38 patients with high myopia (HM) (17 males and 21 females), whose binocular refractive diopter were -6.00 to -7.00 D, and 38 healthy controls (17 males and 21 females), closely matched in age, sex, and education levels, participated in the study. Spontaneous brain activities were evaluated using the voxel-wise DC method. The receiver operating characteristic curve was measured to distinguish patients with HM from healthy controls. Correlation analysis was used to explore the relationship between the observed mean DC values of the different brain areas and the behavioral performance. Compared with healthy controls, HM patients had significantly decreased DC values in the right inferior frontal gyrus/insula, right middle frontal gyrus, and right supramarginal/inferior parietal lobule (P<0.05). In contrast, HM patients had significantly increased DC values in the right cerebellum posterior lobe, left precentral gyrus/postcentral gyrus, and right middle cingulate gyrus (P<0.05). However, no relationship was found between the observed mean DC values of the different brain areas and the behavioral performance (P>0.05). HM is associated with abnormalities in many brain regions, which may indicate the neural mechanisms of HM. The altered DC values may be used as a useful biomarker for the brain activity changes in HM patients.

[Degree centrality of the functional network in schizophrenia patients].

The aim of the present study was to investigate the alternations of brain functional networks at resting state in the schizophrenia (SCH) patients using voxel-wise degree centrality (DC) method. The resting-state functional magnetic resonance imaging (rfMRI) data were collected from 41 SCH patients and 41 matched healthy control subjects and then analyzed by voxel-wise DC method. The DC maps between the patient group and the control group were compared using by two sample t test. The correlation analysis was also performed between DC values and clinical symptom and illness duration in SCH group. Results showed that compared with the control group, SCH patients exhibited significantly decreased DC value in primary sensorimotor network, and increased DC value in executive control network. In addition, DC value of the regions with obvious differences between the two groups significantly correlated to Positive and Negative Syndrome Scale (PANSS) scores and illness duration of SCH patients. The study showed the abnormal functional integration in primary sensorimotor network and executive control network in SCH patients.
